چکیده
The taxane-based therapy provides survival benefit in patients with metastatic prostate cancer; however, the average is less than 20 months due to partial taxane-related chemoresistance. Innovative strategies are needed overcome chemoresistance for improved patient survival. In this project, paclitaxel-resistance was developed on androgen-independent PC3 and androgen-dependent 22Rv1 LNCaP human cancer (PCa) cell lines investigate efficacy of methyl jasmonate (MeJa), an anti-cancer drug, overcoming PCa were maintained RPMI-1640 medium supplemented 10% fetal bovine serum (FBS) at 37?C under 5% CO2. exposed gradually increasing doses paclitaxel. Since resistance could not be achieved, study continued line. It demonstrated that paclitaxel-resistant overexpress ABCB1. Resistance levels cells MeJa activity all resistant parental measured using CellTiter-Glo? luminescent assay. Test results compared Student?s t-test or analysis variance (ANOVA). P?0.05 (two-tailed) considered significant. conclusion, showed more cytotoxicity paclitaxel (PC3-PtxR) (22Rv1-PtxR) cells. Detection cytotoxic effects may contribute development alternative new compounds prevention chemosensitization chemotherapeutics such as

Synergistic Anticancer Effect of Paclitaxel and Noscapine on Human Prostate Cancer Cell Lines
Paclitaxel is the one of the most common chemotherapeutic drugs used for the treatment of prostate cancer. However, its current clinical utility has been limited due to numerous serious side effects and drug resistance. Noscapine is an antitussive opium alkaloid that showed antitumor activity against a variety of cancer while has not exhibited severe side effects. This study investigates the a...

متن کاملThe establishment of two paclitaxel-resistant prostate cancer cell lines and the mechanisms of paclitaxel resistance with two cell lines.
BACKGROUND Although paclitaxel is used for hormone-resistant prostate cancer, relapse definitely occurs later. Details of the molecular mechanism responsible for paclitaxel- resistance remain unclear. METHODS We established paclitaxel-resistant cells, DU145-TxR and PC-3-TxR from parent DU145 and PC-3. To characterize these cells, we examined cross-resistance to other anticancer drugs. Express...
متن کاملAnticancer Effects of Deoxypodophyllotoxin and Juniperus communis L. on Prostate Cancer Cell Lines
Background and purpose: In this study, the effects of a mixture of deoxypodophyllotoxin/DPT and Juniperus communis L. on apoptosis and cellular inhibition were evaluated. Also, their cytotoxicity effects on prostate cancer cells (PC3 and DU145) and normal cells (HGFs), their anti-inflammatory effects, oxidation properties, and their effects on the expression of androgen receptors (AR) and clust...
متن کاملAn AKT activity threshold regulates androgen-dependent and androgen-independent PSA expression in prostate cancer cell lines.
The androgen receptor (AR) plays an important role in early prostate cancer by activating transcription of a number of genes participating in cell proliferation and growth and cancer progression. However, as the cancer progresses, prostate cancer cells transform from an androgen-dependent to an androgen-independent state. Androgen-independent prostate cancer can manifest itself in several forms...
